从新疆地区土壤中经驯化、分离纯化得到一株极端嗜盐菌,并对其形态特征、生理生化特性及16S rDNA同源性进行了检索比较,鉴定该菌株为伸长盐单胞菌(Halomonas elogata);其次,考察了不同因素对菌株降解偶氮染料--活性兰BRF的影响,比较了不同盐度条件下的降解率,并在30℃、5%盐度生长条件下采用单因素实验考察了该菌接菌量、染料初始浓度、初始pH等因素对活性兰BRF72h降解率的影响,采用正交试验L9(33)获得适宜的降解条件;最后,通过紫外-可见吸收光谱分析了降解产物.结果表明,该菌株耐盐范围为0~32%,最适生长盐度为5%~20%;在盐度为3%~25%条件下该菌株对活性兰BRF的降解率随着盐度的升高先升后降,5%盐度下的降解效果最佳;在30℃、5%盐度下适宜降解条件为:染料浓度100mg·L-1,pH=7,接菌量5%,72h的降解率为88.62%.紫外-可见光谱结果显示,降解产物的光谱与活性兰BRF的明显不同,可见光区的吸收峰消失,正己烷萃取液无紫外吸收,二氯甲烷萃取液出现两个紫外吸收峰,推测为芳胺类及杂环类等极性化合物;质谱分析证实,降解产物合苯甲基二胺、2,6-氨基苯酚等极性化合物.  相似文献

The U.S. Renewable Fuel Standard calls for 136 billion liters of renewable fuels production by 2022. Switchgrass (Panicum virgatum L.) has emerged as a leading candidate to be developed as a bioenergy feedstock. To reach biofuel production goals in a sustainable manner, more information is needed to characterize potential production rates of switchgrass. We used switchgrass yield data and general additive models (GAMs) to model lowland and upland switchgrass yield as nonlinear functions of climate and environmental variables. We used the GAMs and a 39-year climate dataset to assess the spatio-temporal variability in switchgrass yield due to climate variables alone. Variables associated with fertilizer application, genetics, precipitation, and management practices were the most important for explaining variability in switchgrass yield. The relationship of switchgrass yield with climate variables was different for upland than lowland cultivars. The spatio-temporal analysis showed that considerable variability in switchgrass yields can occur due to climate variables alone. The highest switchgrass yields with the lowest variability occurred primarily in the Corn Belt region, suggesting that prime cropland regions are the best suited for a constant and high switchgrass biomass yield. Given that much lignocellulosic feedstock production will likely occur in regions with less suitable climates for agriculture, interannual variability in yields should be expected and incorporated into operational planning.  相似文献

The southern pine bark beetle guild (SPBG) is arguably the most destructive group of forest insects in the southeastern USA. This guild contains five species of bark beetles (Coleoptera: Curculionidae: Scolytinae): Dendroctonus frontalis, Dendroctonus terebrans, Ips avulsus, Ips calligraphus, and Ips grandicollis. A diverse community of illicit receivers is attracted to pheromones emitted by the SPBG, including the woodborers Monochamus carolinensis and Monochamus titillator (Coleoptera: Cerambycidae). These woodborers have been traditionally classified as resource competitors; however, laboratory assays suggest that larval M. carolinensis may be facultative intraguild predators of SPBG larvae. This study used polymerase chain reaction (PCR)-based molecular gut content analyses to characterize subcortical interactions between M. titillator and members of the SPBG. The half-lives of SPBG DNA were estimated in the laboratory prior to examining these interactions in the field. A total of 271 field-collected M. titillator larvae were analyzed and 26 (9.6?%) tested positive for DNA of members of the SPBG. Of these larvae, 25 (96.2?%) tested positive for I. grandicollis and one (3.8?%) for I. calligraphus. Failure to detect D. terebrans and D. frontalis was likely due to their absence in the field. I. avulsus was present, but primers developed using adult tissues failed to amplify larval tissue. Results from this study support the hypothesis that larval Monochamus spp. are facultative intraguild predators of bark beetle larvae. Additionally, this study demonstrates the capabilities of PCR in elucidating the interactions of cryptic forest insects and provides a tool to better understand mechanisms driving southern pine beetle guild population fluctuations.  相似文献

为了更好地了解和评价湖南大米中As、Pb和Cd含量及其对人体的健康影响,在对湖南矿区和冶炼区水稻土壤重金属污染调查的基础上,分别以湖南各地市场大米和污染区当地生产的稻谷样品为例,对其进行重金属含量分析及对人体的健康风险评价.结果表明,湖南各地市场大米样品中As、Pb和Cd的平均含量分别是0.20、0.20和0.28mg·kg-1,其中,衡阳市场大米中的As、Pb和Cd含量最高,其次是株洲和湘潭市场的大米.污染区稻谷中As、Pb和Cd含量分布均为:谷壳糙米精米,污染区精米中As、Pb和Cd的含量分别是0.24、0.21和0.65mg·kg-1,其中,来自衡阳常宁市水口山铅/锌矿区的稻谷样品中的As、Pb和Cd含量最高,其次是株洲清水塘冶炼区和湘潭锰矿区的稻谷.与市场大米样品相比,污染区精米中As、Pb和Cd的平均含量比市场大米样品高.无论是市场大米样品,还是从污染区稻田采集的稻米,均以衡阳地区稻米中的As、Pb和Cd污染最为严重,其次为株洲和湘潭地区.在As、Pb和Cd的健康风险评价中,Cd是湖南各地稻米中影响人体健康的主要因子,株洲和湘潭的Cd污染区达到90%以上,其次是As和Pb.  相似文献

基于16S rDNA的PCR-DGGE(polymerase chain reaction-denaturing gradient gel electrophoresis)技术研究了倒置A2/O(anoxic-anaerobic-aerobic)工艺处理染织废水过程中的微生物群落结构.从兼氧、厌氧和好氧区共取6个不同处理阶段的样品,测量了不同处理阶段样品的COD、pH、NH4+-N、H2S、总磷(TP)、污泥含量(TS)、色度及其脱除率,并分析了这些指标的变化规律.结果显示,COD在处理后期降到了145mg·L-1,NH4+-N的脱除率可达到85%,硫化氢的去除率达到了90%以上,磷的脱除率可达80%以上,TS处于逐渐升高的趋势,色度的脱除率达到65%.DGGE图谱显示,好氧处理阶段的微生物多样性明显比兼氧处理和厌氧处理的阶段的要丰富.  相似文献

应用显微与亚显微技术和琼脂糖凝胶电泳方法,研究了不同浓度Cd2+(0、7.25、14.50、29.00、58.00和116.00mg·L-1)处理不同时间(24、48、72和96h)对长江华溪蟹(Sinopotamon yangtsekiense)肝胰腺细胞凋亡的影响.结果显示,29.00mg·L-1Cd2+处理48h,光镜下肝胰腺细胞凋亡主要表现为染色质不规则凝聚、固缩和边集;58.00mg·L-1Cd2+处理72h,细胞核碎裂,形成凋亡小体;116mg·L-1Cd2+处理96h,出现坏死细胞;电镜下48h和72h处理组肝胰腺细胞呈现典型的细胞凋亡特征,与光镜检测结果一致,具体表现为核边集、核膜折叠、核裂解形成凋亡小体.琼脂糖凝胶电泳图谱中48和72h处理组出现凋亡细胞所特有的DNA梯状条带,58.00mg·L-1Cd2+处理72h后条带尤为清晰.随着Cd2+浓度的增加和处理时间的延长,肝胰腺中凋亡细胞所占比例呈现先升后降的趋势,凋亡指数的变化表现出显著的剂量和时间效应关系.研究表明,镉能够诱导长江华溪蟹肝胰腺出现细胞凋亡.细胞凋亡的检测及凋亡指数的变化能够灵敏地反映出镉对水生动物的胁迫程度及毒性大小,可作为水生态环境镉污染的生物评估指标.  相似文献

Over 14 million hectares of erosion prone cropland in the United States has been converted into grasslands through the Conservation Reserve Program (CRP) administered by the United States Department of Agriculture, however, studies of the effects of CRP enrollment on plant communities and subsequent plant succession are largely lacking. In Delta Junction, Alaska plant communities in CRP fields are transitioning from grasslands to shrub dominated plant communities, which are resulting in compliance problems with program regulations that state “fields must be maintained in a condition that permits easy conversion to cropland”. To determine plant succession and how previous land management and soils might influence the transition, we measured plant populations in 20 CRP fields throughout Delta Junction using modified-Whittaker plots. These data were combined with data on current management practices, previous farming history, soils, soil properties, diversity indices, and time since land was cleared and analyzed with nonmetric multidimensional scaling ordination to determine factors that influence plant succession. Time in the CRP was the only factor consistently influencing plant succession. As time in the CRP increased, the planted introduced grasses brome grass (Bromus inermis) and red fescue (Festuca rubra) and the native pteridophyte (Equisetum arvense) decreased, whereas a native grass (Calamigrostis canadensis), five native forb, two native shrub, and three native tree species increased. Plant diversity increased at a rate of more than 2 species per 1000 m2 per year. Regression analyses of plant species and plant groups using time in the CRP as the dependent variable resulted in the identification of outlier CRP fields with significantly more or less than expected covers of vegetation. All fields with these outliers had reasonable explanations for the differences in cover that were unrelated to the overall rate of plant succession. Current management practices will result in incompliant fields and different management practices that result in woody vegetation control is key to maintaining CRP fields in compliance.  相似文献

Reducing carbon emissions from deforestation and degradation in developing countries is of the central importance in efforts to combat climate change. A study was conducted to measure carbon stocks in various land-use systems including forms and reliably estimates the impact of land use on carbon (C) stocks in the forest of Rajasthan, western India (23°3′–30°12′N longitude and 69°30′–78°17′E). 22.8% of India is forested and 0.04% is the deforestation rate of India. In Indian forest sector of western India of Aravally mountain range covered large area of deciduous forest and it’s very helpful in carbon sequestration at global level. The carbon stocks of forest, plantation (reforestation) and agricultural land in aboveground, soil organic and fine root within forest were estimated through field data collection. Results revealed that the amount of total carbon stock of forests (533.64?±?37.54 Mg·ha?1, simplified expression of Mg (carbon) ·ha?1) was significantly greater (P?<?0.05) than the plantation (324.37?±?15.0 Mg·ha?1) and the agricultural land (120.50?±?2.17 Mg·ha?1). Soil organic carbon in the forests (172.84?±?3.78 Mg·ha?1) was also significantly greater (P?<?0.05) than the plantation (153.20?±?7.48 Mg·ha?1) and the agricultural land (108.71?±?1.68 Mg·ha?1). The differences in carbon stocks across land-use types are the primary consequence of variations in the vegetation biomass and the soil organic matter. Fine root carbon was a small fraction of carbon stocks in all land-use types. Most of the soil organic carbon and fine root carbon content was found in the upper 30-cm layer and decreased with soil depth. The aboveground carbon (ABGC): soil organic carbon (SOC): fine root carbon ratios (FRC), was 8:4:1, 4:5:1, and 3:37:1 for the forest, plantation and agricultural land, respectively. These results indicate that a relatively large proportion of the C loss is due to forest conversion to agricultural land.  相似文献

田间微区试验研究了不同种植密度对伴矿景天生长和地上部重金属吸收量的影响.结果表明,适度增大种植密度可促进伴矿景天的生长,显著提高其地上部生物量,但过分密植对植物地上部增产无显著贡献.种植密度由11万株/hm2上升到44万株/hm2时,伴矿景天地上部Cd、Zn吸取量显著上升,分别由0.208 kg.hm-2上升至0.631 kg.hm-2、13.2 kg.hm-2上升至58.7kg.hm-2;但种植密度从44万株/hm2增大到100万株/hm2时,植物地上部重金属吸取量并无显著提高.伴矿景天种植密度为44万株/hm2时,在Cd、Zn分别为(3.04±0.11)mg.kg-1和(1 299±96)mg.kg-1的污染土壤上种植1 a,对镉锌的修复效率分别达21.1%和4.60%,表明选择适宜的密度种植伴矿景天有利于增大植物地上部Cd、Zn吸取量,从而缩短修复时限.  相似文献
